    Product Description

    The PathScan® CD11b/ITGAM Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of CD11b/ITGAM protein. A CD11b/ITGAM rabbit antibody has been coated onto the microwells. After incubation with cell lysates, CD11b/ITGAM protein is captured by the coated antibody. Following extensive washing, a CD11b/ITGAM mouse detection antibody is added to detect the captured CD11b/ITGAM protein. Anti-mouse IgG, HRP-linked Antibody is then used to recognize the bound detection antibody. HRP substrate, TMB, is added to develop color. The magnitude of the absorbance for this developed color is proportional to the quantity of CD11b/ITGAM protein.

    Specificity / Sensitivity

    The PathScan® CD11b/ITGAM Sandwich ELISA Kit detects endogenous levels of CD11b/ITGAM protein. The kit sensitivity is shown in Figure 1. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species.

    Background

    Cluster of differentiation molecule 11b (CD11b)/Integrin alpha M (ITGAM) is a transmembrane protein forming heterodimers that are composed of α and β subunits (1). CD11b is expressed by, and commonly used as a marker for, myeloid lineage cells, including neutrophils, monocytes, macrophages, and microglia (2). CD11b is phosphorylated at Ser1126 (cytoplasmic tail) in neutrophils. Research has shown that this phosphorylation event plays a role for leukocytes traveling from the bloodstream to tissues (3). Furthermore, genome-wide association studies have linked CD11b to autoimmune diseases, such as systemic lupus erythematosus (SLE) (4).
