    Background

    Vesicular monoamine transporters (VMATs) belong to the SLC18 family, a subset of the major facilitator superfamily (MFS). VMAT1, encoded by the SLC18A1 gene, is a proton-dependent pump that sequesters monoamine neurotransmitters, such as dopamine, serotonin, and norepinephrine, from the cytoplasm into synaptic and endocrine secretory vesicles. While VMAT1 is primarily expressed in the peripheral and sympathetic nervous systems, adrenal chromaffin cells, and the enteric endocrine system, it plays a role in regulating stress responses, blood pressure, and neurobehavioral traits (1,2). Conversely, the VMAT2 isoform (SLC18A2) is expressed predominantly in the brain, though both isoforms serve as markers for serotonergic and immature dopaminergic neurons (3,4). Notably, genetic variants of SLC18A1 have been linked to psychiatric conditions, including schizophrenia, bipolar disorder, autism, and anxiety (5,6,7).
