    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu495 of human SF3A3 protein. Antibodies are purified by peptide affinity chromatography.

    Background

    The mammalian splicing factor SF3a contains three primary subunits: SF3A1 (SF3a120), SF3A2 (SF3a66), and SF3A3 (SF3a60), which, along with SF3b, activate the U2 snRNP to form the pre-mRNA splicing complex (1,2). SF3A2 and SF3A3 interact with SF3A1, but not with each other, and SF3A3 plays a major role in recruiting SF3A1 to the complex (3). Each protein in the heterotrimer has been shown to be essential for mRNA splicing and cell viability (4). Notably, SF3a plays a role in growth factor signaling as SF3A1 can bind to ERα when phosphorylated at Ser118 to affect downstream splicing (5). SF3a also plays a role in the innate immune response, promoting inflammation by splicing the TLR adaptor Myd88. Furthermore, SF3a regulates several genes involved in innate immune signaling pathways in macrophages versus other pathways (6,7).
