    Background

    Thymocyte-expressed molecule involved in selection protein 2 (THEMIS2) is a member of the THEMIS family of proteins, initially identified as a scaffold protein involved in TLR signaling and tumor cell differentiation (1). THEMIS2 contains two conserved cysteine-based domains, a proline-rich region, and a nuclear localization signal (1-4). LPS stimulation triggers THEMIS2 phosphorylation and activation, facilitating its interaction with several key MAP kinase proteins and action in several immune cell types (1). THEMIS2 is a key player in T cell development following TCR engagement (2-4). In natural killer (NK) cells, THEMIS2 regulates memory function and formation, and can regulate antitumor activity by inhibiting receptors that activate NK cell signaling (2,3). THEMIS2 is involved in the positive selection of B cells and germinal center B cells by self and foreign antigens. THEMIS2 has been shown to lower the threshold for B cell activation by low-avidity, but not high-avidity, antigens (4). Recent research has focused on the role of THEMIS2 in cancer biology (5-8). Its transcription is estrogen-responsive, and THEMIS2 protein expression is implicated in breast and ovarian cancers (6-8). Elevated THEMIS2 protein levels are seen in ovarian and endometrial carcinomas (7,8). Significantly increased THEMIS2 expression was shown in breast cancer cells (8), underscoring its potential as a target for cancer therapies.
