    Background

    Dead box protein 1 (DDX1) is a DEAD box RNA helicase that is involved in 3’-end processing of pre-mRNA. DDX1 is capable of binding to poly(A) RNA and interacts with several key RNA-binding proteins, including hnRNP K, and forms discrete nuclear foci with the CSTF complex called DDX1 bodies (1,2). During DNA damage, DDX1 is recruited to double-strand breaks (DSBs) upon phosphorylation by ATM, where it plays a role in RNA clearance at the DSB sites (3). DDX1 phosphorylation has also been shown to increase expression of a subset of microRNAs (miRNAs) that can inhibit tumor progression (4). DDX1 has been shown to bind to the HIV-1 Rev protein, and DDX1 downregulation disrupts viral production (5,6). Additionally, DDX1 has been implicated in other viral infections, including SARS-CoV-2, where DDX1 binds the nucleocapsid protein to inhibit its function (7). DDX1 has been described in various cancer types, but notably, the DDX1 gene is co-amplified with MYCN in neuroblastoma (8-10).
