    Background

    Tripartite motif-containing (TRIM) proteins constitute a distinct family, many of which function as ubiquitin E3 ligases that mediate the degradation of normal regulatory proteins. Emerging evidence suggests that TRIMs can also promote the degradation of aberrant proteins, a property attributable to small ubiquitin-like (UBL) modifier (SUMO) E3 ligase activity (1). TRIM2 is a member of TRIM class VII, a class defined by the presence of a C-terminal NHL domain (2). It is predominantly expressed in the brain, where it is present in the cytoplasm and in cytoplasmic filaments, and has been reported to enhance axon specification during neuronal polarization (3). More recently, TRIM2 has also been implicated in regulating intracellular lipid levels, protecting cells from glutamine deprivation-induced apoptosis, and playing an antiviral role by limiting New World arenavirus entry into cells (4,5).
