    Background

    DNA lesions in the template strand of actively transcribed genes result in genome-wide transcriptional arrest. The transcription-coupled nucleotide excision repair (TC-NER) pathway efficiently eliminates transcription-blocking DNA lesions (TBLs) (1). ELOF1 is a component of the transcriptional elongating Pol II complex (2). The protein ensures high-speed elongation processivity during mRNA transcription on the DNA template (3). ELOF1 is an important player of the TC-NER-complex (4). ELOF1 facilitates Pol II ubiquitylation and ensures correct TC-NER-complex assembly upon DNA damage (5). The protein promotes UV-stimulated scaffold protein A (UVSSA) and lesion-stalled Pol II binding, resulting in subsequent transcription factor IIH (TFIIH) recruitment, which is crucial in the full incision complex assembly to excise the TBL and restart transcription (6).
