    Background

    Phosphatidylinositol 4-kinase beta (PI4KB) is an enzyme that phosphorylates phosphatidylinositol (PI) to produce phosphatidylinositol 4-phosphate (PI4P), a key lipid involved in membrane trafficking and lipid metabolism (1). PI4KB is predominantly localized to the Golgi apparatus, where it is recruited by the Golgi resident protein acyl-CoA-binding domain-containing 3 (ACBD3) (2). PI4P secreted at the Golgi apparatus and trans-Golgi network (TGN) is critical for the recruitment of effector proteins that regulate vesicle formation and lipid transport (3). PI4KB is also essential for the replication of many RNA viruses, including hepatitis C virus (HCV) and Aichi virus (AiV), as it facilitates the formation of replication organelles by generating PI4P-enriched membranes (4,5). As a result, several PI4KB inhibitors have been designed as potential antiviral therapeutics (6,7).
