    Background

    Zinc and RING finger 3 (ZNRF3) is a transmembrane E3 ubiquitin ligase containing several domains, including an extracellular PA domain, an R-spondin (RSPO) binding domain, and a RING-type E3 ligase domain (RING) (1). ZNRF3 is associated with the Wnt receptor complex on the cell surface and inhibits Wnt signaling by promoting the turnover of Frizzled. RSPO interacts with the extracellular domain of ZNRF3 and induces the association between ZNRF3 and LGR4/5/6, which results in the membrane clearance of ZNRF3 (2-4). Under normal physiological conditions, ZNRF3 contributes to adrenal homeostasis (5), liver homeostasis and regeneration (6), neurodevelopment (7), and sex determination (8). ZNRF3 functions as a tumor suppressor in various cancer types, including osteosarcoma, gastric cancer, and papillary thyroid and nasopharyngeal carcinomas (9-12). ZNRF3 downregulation promotes Wnt signaling and cancer cell proliferation and invasion. In prostate cancers, ZNRF3 loss is a predictor of metastatic relapse (13).
