    Background

    B cell scaffold protein with ankyrin repeats (BANK1) is an adaptor protein that is highly expressed in B lineage but not T lineage cells (1). BANK1 is phosphorylated upon B cell antigen receptor (BCR) stimulation and functions as a scaffolding protein, promoting Lyn-mediated phosphorylation of inositol 1,4,5-triphosphate receptor (IP3R) and downstream calcium signaling (1). BANK1 also associates with B lymphoid tyrosine kinase (BLK) and phospholipase C gamma 2 (PLCg2), two additional mediators of B cell signaling (2,3). Knockout studies in mice suggest that BANK1 additionally acts as a negative regulator of CD40-mediated Akt activation (4). BANK1 gene mutations are associated with several diseases, including systemic lupus erythematosus (SLE), systemic sclerosis, and rheumatoid arthritis (5-7).
