    Background

    RUN and FYVE domain-containing protein 3 (RUFY3) is an actin-binding protein that is enriched in the brain. RUFY3 was identified as a protein that is upregulated during neuronal polarization, which is important for proper neuron functioning (1). RUFY3 likely functions in regulating neuronal polarization by interacting with the neuronal cytoskeleton via actin filament-binding proteins, such as Fascin (2). RUFY3 undergoes various forms of posttranslational modifications, including CASP3-dependent cleavage and phosphorylation, which may modulate selective synaptic and axonal degeneration during brain development and neurodegenerative disease (3).
