    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding His300 of human 5-HT2A receptor protein.

    Background

    Serotonin (5-hydroxytryptamine, 5-HT) is a monoamine neurotransmitter abundant in the central nervous system. Seven distinct 5-HT receptor subtypes (5-HT1-7) have been identified. Most belong to the G protein-coupled receptor (GPCR) superfamily, which is characterized by seven transmembrane domains and intracellular signaling cascades, including cyclic AMP production (1,2). Within the 5-HT2 family, three distinct genes encode the 5-HT2A, 5-HT2B, and 5-HT2C receptors (3). The 5-HT2A receptor is broadly distributed throughout the brain, with enrichment in the dendrites of layer IV and V cortical pyramidal neurons (4). As the principal receptor for serotonin, 5-HT2A modulates cognition, mood, and endocrine regulation, and its dysfunction is implicated in several psychiatric disorders (5). 5-HT2A has been recognized as a primary target for atypical antipsychotics, certain antidepressants, and anxiolytic agents. Its activation by the serotonergic psychedelic psilocybin is being explored as a therapeutic option for treatment-resistant depression (6,7).
