    This product is the carrier free version of product #10162. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

    This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

    BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity.

    Formulation

    Supplied in 1X PBS (10 mM Na2HPO4, 3 mM KCl, 2 mM KH2PO4, and 140 mM NaCl (pH 7.8)). BSA and Azide Free.

    Storage

    Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

    Background

    Non-POU domain-containing octamer-binding protein (NONO), also referred to as p54nrb, is a DNA and RNA-binding protein that is involved in several nuclear processes. NONO binds to the conventional octamer duplex of double-stranded DNA, and can additionally bind to single-stranded DNA and RNA at a location independent of the duplex site (1). NONO belongs to the Drosophila melanogaster behavior, human splicing (DBHS) protein family along with paraspeckle protein 1 (PSPC1) and splicing factor, proline- and glutamine-rich (PSF/SFPQ) (2). NONO, PSPC1, and PSF/SFPQ associate together along with long nonprotein-coding RNA NEAT1 to form ribonucleic protein bodies referred to as paraspeckles, which play a role in gene expression and regulation (2). NONO heterodimerizes with PSF/SFPQ to process pri-miRNA prior to presentation to the Drosha complex (3). The NONO/PSF heterodimer can also stabilize paired DNA ends and promote DNA double-strand break repair by canonical non-homologous end rejoining (4). NONO also forms a complex with PSF/SFPQ and DExH-box helicase 9 (DHX9) that produces a long isoform of BIN1, promoting carcinogenesis in hepatocellular carcinoma (5). In breast cancer, NONO regulates pre-mRNA splicing and binding to mRNAs of cell proliferation-related genes, regardless of hormone sensitivity (6). NONO has also been hypothesized to play a role in heart disease, and a recent study in mice showed that NONO deficiency led to heart defects by fibroblast hyperproliferation (7).
