    Background

    Hydroxycarboxylic acid receptor 2 (HCAR2), also known as G protein–coupled receptor 109A (GPR109A), is widely expressed in white and brown adipose tissue, keratinocytes, and various immune cells, including monocytes, macrophages, neutrophils, dendritic cells, and microglia (1,2). HCAR2/GPR109A has three significant ligands: nicotinic acid (NA), sodium butyrate (SB), and β-hydroxybutyric acid (BHBA) (2,3). HCAR2/GPR109A activation triggers robust anti-inflammatory effects, leading to anti-atherogenic properties and displaying a role as a potential therapeutic target for treatment of many inflammatory diseases, including neuroinflammatory and/or neurodegenerative diseases such as those affecting the retina and Alzheimer’s disease (2,4,5).
