    Background

    Zinc finger DHHC domain-containing protein 8 (ZDHHC8) is a palmitoyltransferase highly expressed within the central nervous system, localizing to the Golgi, endoplasmic reticulum (ER), or plasma membrane. As a protein S-acyltransferase, it contains conserved DHHC (aspartate-histidine-histidine-cysteine) tetrapeptides, zinc finger structural domains, as well as four transmembrane domains (1,2). ZDHHC8 attaches palmitate to protein substrates, regulating their membrane localization, stability, and trafficking. It plays critical roles in brain development, synaptic plasticity, and has been associated with schizophrenia susceptibility and epilepsy (3,4). It also regulates cholesterol transport, SARS-CoV-2 spike protein processing, and ferroptosis resistance in cancer cells (5).
