    Monoclonal antibody is produced by immunizing animals with a recombinant protein specific to the N-terminus of human ROR gamma protein. This antibody recognizes residues surrounding Gln142 of human ROR gamma protein.

    Background

    RAR-related orphan receptor gamma (RORγ) is encoded by the RORC gene, and belongs to the nuclear receptor family of transcription factors (1). Two isoforms of RORγ have been described: the canonical isoform (also known as RORγ1), and a shorter isoform (known as RORγt or RORγ2). The messenger RNAs for both are transcribed from the RORC gene, but alternative exon usage yields the two distinct isoforms, which differ in the amino-terminal region. Abundant expression of mRNA encoding RORγ1 is detected in multiple tissue types, but little evidence exists to support RORy1 expression at the protein level. By contrast, expression of RORγt protein is readily detected, but appears to be restricted exclusively to distinct subsets of thymocytes and their derivatives (2). Functional studies in mice and humans have revealed that RORγt functions as a master regulator that coordinates immune cell development at multiple stages (3). For example, isoform-specific gene knockout studies in mice have shown that RORγt specifically is indispensable for fetal development of lymph nodes and Peyer's patches (3,4). Postnatally, RORγt functions as a critical regulator of Th17 cell lineage commitment, suggesting that RORγt may have potential as a therapeutic target in inflammatory diseases (5).
