    Background

    NVP-DKY709 is a selective cereblon (CRBN) glue degrader that targets the IKZF2 Helios (Ikaros family zinc finger 2, IKZF2) protein for degradation. Helios is a member of the Ikaros transcription factor family that suppresses immune system response to pathogens by promoting regulatory T cell (Treg) activity (1). Treg cells are responsible for suppressing excessive immune responses and exhausting tumor-specific effector T (Teff) cell actions (1), inhibiting immune responses to cancer cells (2). Glue degraders on the market and in trial phases degrade other members of the Ikaros transcription factor family, including IKZF1 & IKZF3, which is suboptimal due to potential increase in immunosuppressive actions (1); NVP-DKY709 has been found only to degrade IKZF2, leaving IKZF1/3 untouched (3), and has a DC50 of 4 nM in cellular assays (1).
