    Pomalidomide is supplied as a lyophilized powder. For a 15 mM stock, reconstitute 5 mg of powder in 1.22 mL of DMSO. Working concentrations and length of treatment can vary depending on the desired effect. Caution: Teratogenic. May cause harm to the unborn child. Avoid all contact with this product during pregnancy.

    Storage

    Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 1 month to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

    Background

    Pomalidomide, a derivative of thalidomide, is an immunomodulatory drug (IMiD) that binds to the CRL4 E3 ubiquitin ligase substrate receptor, cereblon (CRBN), leading to the targeted degradation of transcription factors Ikaros and Aiolos (1,2). The degradation of these transcription factors initiates several downstream responses that play a role in pomalidomide’s antitumor and immunomodulatory properties (3). Pomalidomide enhances T cell and natural killer (NK) cell-mediated responses, resulting in potent anti-inflammatory and anti-tumor effects while inhibiting the expression of pro-inflammatory cytokines, such as tumor necrosis factor–alpha (TNF-α) and interleukin-6 (IL-6) (3,4).

    Pomalidomide is FDA-approved and widely used in combination therapies for multiple myeloma (MM), particularly in patients who have relapsed or become refractory to other treatments (4). When taken in combination with low-dose dexamethasone, pomalidomide has demonstrated efficacy in reducing disease progression and improving survival rates for patients with MM (4).
