Sildenafil Citrate is supplied as a lyophilized powder. For a 10 mM stock, reconstitute 5 mg of powder in 0.75 mL of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.

Storage

Store lyophilized at room temperature, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 3 months to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

Background

Sildenafil Citrate is an inhibitor of phosphodiesterase 5 (PDE5), which catalyzes the hydrolysis of cyclic guanosine monophosphate (cGMP) in regulating cellular responses to a number of extracellular signals. Sildenafil Citrate binds the catalytic site of PDE5 (Ki = 1 nM) more strongly than cGMP (Ki = 200 nM) (1). Because the PDE5 isozyme is prominently expressed in the corpus cavernosum and the cardiovascular system, PDE5 inhibitors, such as Sildenafil Citrate, have been used therapeutically to treat erectile dysfunction and pulmonary arterial hypertension (2,3). Sildenafil Citrate treatment of a porcine model of cardiac arrest and resuscitation produced a cardioprotective effect, improving cardiac function and survival while reducing apoptosis (4). Through controlling PDE5-mediated cGMP concentrations, Sildenafil Citrate can attenuate the biological effects of nitric oxide (NO) signaling, and play a role in regulating cerebral blood flow and protection against brain injury pathogenesis. Preclinical and clinical trials have studied the potential role of Sildenafil Citrate to improve recovery after brain injury in both adults and neonates (5).
