    Sirtinol is supplied as a lyophilized powder. For a 10 mM stock, reconstitute 5 mg of powder in 1.27 mL of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.

    Storage

    Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 1 month to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

    Background

    Sirtinol is a cell-permeable inhibitor of sirtuin NAD+-dependent histone deacetylases (1). It inhibits yeast sirtuin Sir2p with an IC50 value of 68 μM, and inhibits human SIRT1 and SIRT2 with IC50 values of 131 μM and 38 μM, respectively (1,2). Sirtinol does not inhibit human HDAC1 activity (1). It elevates endogenous levels of reactive oxygen species to induce apoptosis in cancer cells (3). Sirtinol inhibits proliferation of non-small cell lung cancer cells and suppresses inflammatory activity in human dermal microvascular endothelial cells (3,4). Sirtinol also activates the auxin signaling transduction pathway in Arabidopsis, altering plant growth and development (5).
