    Storage

    Background

    Stimulator of interferon genes (STING) Degrader-1 is a compound known as a proteolysis-targeting chimera (PROTAC) with a DC50 of 3.2 μM (1). The small molecules are heterobifunctional, consisting of two protein binding parts joined by a linker. One part of the molecule binds a protein of interest (POI), while the other part binds an E3 ubiquitin ligase, bringing the E3 ligase close to the POI for ubiquitination-coupled protein degradation (2). This complex tags the protein of interest for degradation by the proteasome, decreasing the levels of STING in a cell (3). STING protein is involved in the innate immune response through the activation of type I interferons in response to infections, and can also be linked to autoimmune disorders due to excessive activation. STING Degrader-1 is a potential therapeutic compound used when autoimmune diseases are caused by overactive STING and can lead to excessive immune responses, including chronic inflammation (1).
