    Product Description

    The rapid protocol (RP) PathScan® RP Total Jak2 Sandwich ELISA Kit is a solid-phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of Jak2 protein in a reduced assay time of 1.5 hours. Incubation of cell lysates and detection antibody on the coated microwell plate forms a sandwich with Jak2 in a single step. The plate is then extensively washed, and TMB reagent is added for signal development. The magnitude of absorbance for the developed color is proportional to the quantity of Total Jak2.     The PathScan® RP Total Jak2 Sandwich ELISA Kit detects endogenous levels of Jak2, as shown in Figure 1. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species.

    Background

    Members of the Janus family of tyrosine kinases (Jak1, Jak2, Jak3, and Tyk2) are activated by ligands binding to a number of associated cytokine receptors (1). Upon cytokine receptor activation, Jak proteins become autophosphorylated and phosphorylate their associated receptors to provide multiple binding sites for signaling proteins. These associated signaling proteins, such as Stats (2), Shc (3), insulin receptor substrates (4), and focal adhesion kinase (FAK) (5), typically contain SH2 or other phospho-tyrosine-binding domains.
