    Source / Purification

    Monoclonal antibody is produced by immunizing animals with a recombinant protein fragment specific to the transcriptional activation and negative regulatory domains of human a-Myb protein. The antigen has been further categorized as corresponding to residues surrounding Ala383 of human a-Myb protein.

    Background

    The transcription factor a-Myb, encoded by the gene MYBL1, is a conserved DNA-binding protein that is critical for both spermatogenesis and mammary gland development (1). Like family members b-Myb and c-Myb, a-Myb contains an N-terminal DNA-binding domain that binds the sequence 5'-YAAC[GT]G-3', a trans-activating domain (TAD), and a C-terminal inhibitory domain (2-4). MYBL1 expression is regulated by cell cycle progression to G1/S phase, and a-Myb protein activity is further regulated by cyclin A/CDK2-dependent phosphorylation of the inhibitory domain (5). a-Myb regulates spermatogenic meiosis through transcriptional control of multiple targets, including RFX2 and Sox30 (6,7). a-Myb and c-Myb N-terminal fusions with the NFI-B transcription factor C-terminus are common oncogenic drivers of salivary gland adenoid cystic carcinoma (ACC) (8-11).
