    Background

    Arylsulfatase A (ARSA) is a lysosomal enzyme that hydrolyzes sulfated sphingolipids known as sulfatides (1). ARSA gene mutations lead to sulfatides accumulation in the brain, liver, and kidneys, causing metachromatic leukodystrophy (MLD), a severe neurological disorder that destroys the protective myelin sheath, leading to loss of motor skills and cognitive function (1,2). Beyond its role as a metabolic enzyme, ARSA functions as a molecular chaperone for proteins involved in neurodegeneration, making it a risk gene for Parkinson’s disease (PD) (3,4).
