    The Cereblon Molecular Glue Neosubstrate Profiling Antibody Sampler Kit provides an economical means of detecting proteins known to be targeted as neosubstrates by molecular glues that interact with cereblon. The kit includes enough antibodies to perform two western blot experiments with each primary antibody.

    Background

    CRBN/cereblon, substrate receptor for the CRL4 E3 ubiquitin ligase, has been widely targeted through molecular glue technology to engage neosubstrates that enable targeted protein degradation (1). Thalidomide analogs, also referred to as immunomodulatory imide drugs (IMiDs), target several proteins for degradation, including IKZF1/Ikaros (2, 3), IKZF3/Aiolos (3), CK1α (4,5), Sall4 (6), and eRF3/GSPT1 (7). Ikaros and Aiolos are zinc-finger DNA-binding proteins that belong to the Krüppel transcription superfamily, regulating lymphoid cell development and tumorigenesis (8,9). Casein Kinase I (CK1) is part of a family of protein kinases that consists of multiple isoforms (α, α', β, γ1-3, δ, and ε), having roles in multiple cellular pathways and are considered potential targets in hematological cancers (10). Sall4 is a member of the SALL gene family that encodes putative zinc finger transcription factors highly expressed during development and is a major regulator of pluripotency (11). Eukaryotic release factor 3 (eRF3), also known as G1 to S phase transition 1 (GSPT1), is a GTP-binding protein that regulates G1 to S phase transition and functions in protein translation termination (12).
