    Background

    DnaJ homolog subfamily C member 9 (DNAJC9) serves a dual role as both a histone chaperone and a heat shock co-chaperone. As a histone chaperone, DNAJC9 forms a co-chaperone complex with MCM2 and histone H3-H4 heterodimers, potentially assisting MCM2 in recognizing these heterodimers and facilitating their assembly into nucleosomes. DNAJC9 interacts with HSP70, activating its ATPase activity (1).

    Overexpression of CENP-A, a centromeric histone H3 variant, is frequently observed in cancer. Its mislocalization to noncentromeric regions promotes chromosomal instability (CIN), a hallmark of cancer. DNAJC9 plays a crucial role in preventing the mislocalization of centromeric CENP-A, thereby mitigating chromosome instability (2). DNAJC9 expression is a potential prognostic biomarker in basal-like and luminal A breast cancer (3).
