    For optimal ChIP results, use 10 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.

    Background

    F-box and leucine-rich repeat protein 11 (FBXL11), also known as lysine-specific demethylase 2A (KDM2A) and JmjC domain-containing histone demethylation protein 1A (JHDM1A), is a lysine-specific histone demethylase protein that demethylates Lys4 and Lys36 of histone H3 (1,2). FBXL11 maintains heterochromatin and promotes epigenetic silencing by associating with centromeres and recruiting HP1 (3). Furthermore, FBXL11 is recruited to non-methylated CpG islands by its zinc finger CxxC domain (3). FBXL11 can also control ribosome biogenesis by preventing the transcription of rRNA genes (4). During DNA damage, FBXL11 is phosphorylated by ATM, which reduces its ability to bind to chromatin, allowing H3K36 dimethylation to accumulate and recruit the MRE11 complex (5). Additionally, there is a short isoform of FBXL11 that lacks a JmjC domain and histone demethylation activity. The short isoform has been implicated in breast cancer progression and binds in distinct complexes to HP1alpha (6,7).
