    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly100 of human HMGCL protein.

    Background

    Mitochondrial hydroxymethylglutaryl-CoA lyase (HMGCL) catalyzes the conversion of 3-hydroxy-3-methylglutaryl-CoA to acetyl-CoA and acetoacetate (a ketone body). It is a key enzyme in ketogenesis (1,2). Studies show that HMGCL expression increases in BRAF V600E-expressing human melanoma cells, resulting in upregulated levels of intracellular acetoacetate. Acetoacetate enhances binding of BRAF V600E to MEK1, stimulating activation of the MEK-ERK signaling pathway. Suppression of HMGCL leads to reduced proliferation of these human melanoma cells (2). In addition, HMGCL and β-hydroxybutyrate (a ketone body) are shown to be essential to promote pancreatic ductal adenocarcinoma (PDA) tumor progression (3).
