    Background

    Impaired mitochondrial integrity and function lead to the activation of a transcriptional response known as the mitochondrial unfolded protein response (UPRmt) (1). Mitochondrial 10 kDa heat shock protein (HSP10), also known as heat shock protein family E member 1 (HSPE1), is a mitochondria-specific protein chaperone and a component in the UPRmt pathway. Studies show that Akt3 depletion in primary endothelial cells causes mitochondrial stress and leads to upregulated expression of HSP10 along with HSP60, another mitochondria-specific protein chaperone. This finding suggests a critical role of Akt3 in maintaining mitochondrial homeostasis (2). In addition, HSP10 and HSP60 chaperones were shown to be the most abundant mitochondrial proteins (3).
