    Background

    Interleukin-1 receptor type 1 (IL-1R1) is a transmembrane protein in the interleukin-1 (IL-1) protein family (1-3). IL-1R1 is the receptor for IL-1, which plays a central role in inflammatory responses (1-3). The IL-1 signaling cascade is initiated by the binding of interleukin-1 alpha (IL-1α) or interleukin-1 beta (IL-1β) to IL-1R1, which recruits the IL-1 receptor accessory protein (IL-1RAP) to form a receptor complex, leading to downstream expression of proinflammatory cytokines and chemokines (1-4). The IL-1 receptor antagonist (IL-1Ra) competes with IL-1α and IL-1β to bind IL-1R1, acting as an inhibitor of the IL-1 signaling pathway (4). Aberrant activation of the IL-1 pathway is associated with multiple disorders, such as rheumatoid arthritis, degenerative diseases, cardiovascular disorders, and cancer (4).
