    Background

    Interleukin-21 receptor (IL-21R) is a type I cytokine receptor that forms a heterodimer with the common gamma chain (γc), also known as CD132 (1,2). Upon binding its ligand, IL-21, IL-21R activates downstream targets including Stat1, Stat3, PI3K, and ERK1/2 (2,3). IL-21R is expressed on a variety of lymphoid and myeloid cells, and its expression on T cells can be induced by T cell receptor activation (4-6). IL-21R plays an important role in the differentiation process of several immune subsets, such as follicular helper T cells, Th17 cells, and plasma cells, and helps to activate NK cells (7-10). IL-21R has been implicated in several inflammatory diseases, including Crohn's disease, systemic lupus erythematosus, and rheumatoid arthritis. Hyperactivation of IL-21R signaling drives inflammation (11-14).
