    Background

    The integrator complex is a multiprotein complex consisting of multiple subunits that regulates the termination of RNA polymerase II (RNAPII) transcription (1,2). The integrator was first discovered as a complex that processes the 3′ end of small nuclear RNAs, but subsequent studies have also established the importance of this complex in processing enhancer, viral, and telomeric RNAs (3).

    INTS6 (integrator complex subunit 6), also known as DICE1 (deleted in cancer 1), was first identified in non-small cell lung cancer cells and within the 13q14 chromosomal region (4). Given the susceptibility of this region to allelic deletions in many solid tumors, INTS6 has been extensively examined as a tumor suppressor gene in humans (4-6). This protein was also identified as one of the integrator complex subunits (7). Its primary role is to facilitate protein phosphatase 2A (PP2A) assembly into the integrator-PP2A complex, ultimately contributing to target gene repression (8). Downregulation of INTS6 expression is associated with a poorer prognosis in hepatocellular carcinoma patients (9), while INTS6 levels were found to be significantly increased in colorectal carcinoma tissues and associated with poor prognosis (10).
