    Background

    Leucine-rich repeat-containing protein 31 (LRRC31) belongs to the LRR superfamily, which plays an important role in cell cycle regulation, chromosome stability, apoptosis, and DNA repair (1,2). LRRC31 functions as a tumor suppressor in cancer development. In lung adenocarcinoma (LUAD), β-hydroxybutyrate dehydrogenase 1 (BDH1) promotes LUAD progression through downregulation of LRRC31 expression (3). During breast cancer metastasis to the brain, LRRC31 functions as a DNA repair inhibitor through interaction with Ku70/Ku80 and ATR, sensitizing tumor metastasis to radiation therapy (4). In eosinophilic esophagitis (EoE), IL-13 upregulates LRRC31 expression, promoting epithelial barrier function by downregulating kallikrein expression (5). LRRC31 has also been identified as a novel regulator of pluripotency maintenance in mouse embryonic stem cells (mESCs) by facilitating Stat3 phosphorylation in the Jak/Stat signaling pathway (6).
