    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly155 of human LYPLA1/APT1 protein.

    Background

    Protein palmitoylation and depalmitoylation cycling is suggested to have an important role in diabetes pathophysiology. Lysophospholipase 1 (LYPLA1), also known as acyl-protein thioesterase 1 (APT1), catalyzes protein depalmitoylation. Hyperglycemia was shown to reduce the enzymatic activity of LYPLA1/APT1 in human islets. LYPLA1/APT1 deficiency leads to insulin hypersecretion. Furthermore, LYPLA1/APT1 deficiency induces pancreatic β cell failure in mice fed with a high-fat diet (1). In addition, the autophagy receptor SQSTM1/p62 undergoes reversible S-acylation catalyzed by zinc-finger Asp-His-His-Cys S-acyltransferase 19 (ZDHHC19) and LYPLA1/APT1, respectively. S-acylation of SQSTM1/p62 by ZDHHC19 enhances degradation of ubiquitinated proteins via autophagy. LYPLA1/APT1 deacylates SQSTM1/p62, suppressing SQSTM1/p62-mediated selective autophagy (2,3).
