Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

    Protocol

    Specificity / Sensitivity

MBOAT2 (F8Z4U) Rabbit mAb recognizes endogenous levels of total MBOAT2 protein. This antibody does not cross-react with MBOAT1 protein. Non-specific mitotic staining may be observed by immunofluorescence.

    Species Reactivity:

    Human

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human MBOAT2 protein.

    Background

Membrane-bound glycerophospholipid O-acyltransferase 2 (MBOAT2) is a phospholipid-modifying enzyme. It transfers an acyl group in an acyl-CoA to a lysophospholipid to produce a phospholipid (1,2). Studies show that MBOAT2 is a ferroptosis suppressor, and this function is independent of GPX4 or AIFM2/FSP1. MBOAT2 selectively catalyzes the transfer of monounsaturated fatty acids (MUFAs) to lysophosphatidylethanolamine (lyso-PE), producing more cellular PE-MUFAs and therefore reducing cellular PE-PUFAs (polyunsaturated fatty acids). MBOAT2 suppresses ferroptosis through phospholipid remodeling. In addition, MBOAT2 expression is upregulated by androgen receptor (AR) signaling in AR+ prostate cancer cells, therefore inhibiting their ferroptosis (3).
