    Background

    Macrophage-expressed gene 1 protein (MPEG1), also known as perforin-2, is a member of the Membrane Attack Complex/Perforin (MACPF) superfamily of pore-forming proteins (1,4). MPEG1, mainly expressed in macrophages and leukocytes, can be upregulated in response to proinflammatory signals such as tumor necrosis factor α (TNF-α) and lipopolysaccharides (LPS) (1-3). MPEG1 is involved in both innate and adaptive immunity, localizing to early endosomes and phagosomes/lysosomes, and acts in an antimicrobial capacity (4). In dendritic cells, MPEG1 helps deliver antigens into the cytosol, where they can be processed and presented on MHC class I molecules to activate CD8+ T cells (5).
