    Background

    MPP5 (Membrane Palmitoylated Protein 5)/PALS1 (Protein-Associated with Lin7) belongs to the MAGUK (membrane-associated guanylate kinase) family whose members typically function as molecular scaffolds in the assembly of multiprotein complexes. MAGUK family members contain PSD-95–Discs Large–zona occludens-1 (PDZ), Src homology 3 (SH3), and guanylate kinase (GuK) domains (1). In addition, MPP5/PALS1 contains two L27 domains, L27C and L27N, that bind to a multi-PDZ domain protein called Pals1-associated tight junction (PATJ) (1,2). MPP5/PALS1 is essential for cellular and tissue polarity, epithelial cell adhesion, and the formation of tight junctions (2,3). This scaffolding protein is involved in the development of ocular tissue and localizes in the Golgi of the retinal cells (3). Studies have suggested that disruption of MPP5/PALS1 by SARS-CoV small envelope E protein can lead to deterioration of the lung epithelium in infected individuals (4).
