    Background

    Sialidases, also known as N-acetyl-alpha-neuraminidases, catalyze hydrolytic cleavage of the terminal sialic acid on a glycan of glycolipids and glycoproteins. In mammalian cells, there are four different sialidases: NEU1, NEU2, NEU3, and NEU4. They vary in their subcellular localizations and substrate specificities. These sialidases regulate the function of their target glycolipids and glycoproteins in many biological processes (1,2). Studies show that NEU1 associates with the juxtamembranous segment of the cytoplasmic domain of the membrane-spanning sialoglycoprotein mucin-1 to suppress PI3K-Akt signaling (3). Furthermore, NEU1 plays a role in regulating insulin receptor. Inhibition of the sialidase activity of NEU1 by interfering peptides reduces insulin receptor activity in vitro and glucose homeostasis in vivo (4).
