    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu814 of human NLRP4 protein.

    Background

    NOD-like receptor (NLR) family pyrin domain containing 4 (NLRP4), also known as NALP4, is a cytosolic protein with roles in regulating both autophagy, through interactions with beclin-1, and the type I interferon response by targeting TBK1 for degradation (1,2). NLRP4 also serves as an intracellular sensor for microbes and it is recruited to phagosomes containing group A Streptococcus, where it associates with Rho GTPases and Rho GDP dissociation inhibitor alpha (ARHGDIA) to promote xenophagy (3). NLRP4 may also play a role in cancer, as non-small cell lung cancer patients with NLRP4 mutations displayed improved response to immune checkpoint blockade (4). In mice, NLRP4 knockdown led to enhanced type I interferon production and slower tumor growth (4). Conversely, NLRP4 upregulation enhances DNA damage repair in pancreatic cancer cell lines, and higher NLRP4 expression negatively correlates with mouse BxPC-3 xenograft response to olaparib treatment (5).
