    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val339 of human P5CS/ALDH18A1 protein.

    Background

    Delta-1-pyrroline-5-carboxylate synthase (P5CS), also known as aldehyde dehydrogenase family 18 member A1 (ALDH18A1), catalyzes the rate-limiting step in the biosynthesis of proline and is critical for collagen biosynthesis. This enzyme is therefore important in pathological angiogenesis (1). In addition, P5CS/ALDH18A1 is often upregulated in tumor development. Studies show that it is highly expressed in hepatocellular carcinoma (HCC). P5CS/ALDH18A1 knockdown leads to decreased mitochondrial respiration, suppressing HCC cell proliferation and migration. SIRT2 deacetylates P5CS/ALDH18A1 and activates its enzymatic activity, resulting in increased mitochondrial respiration to support hepatocellular carcinoma growth (2). Moreover, when the demand for oxidative phosphorylation (OXPHOS) increases, P5CS/ALDH18A1 becomes sequestered in a subpopulation of mitochondria lacking cristae and ATP synthase, helping to maintain both OXPHOS and the reductive biosynthesis of proline within the cell (3).
