    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human p67phox protein.

    Background

    p67phox is a component of the phagocytic NADPH oxidase complex, which is composed of membrane-bound p91phox and p22phox heterodimers (also known as cytochrome b558), the cytosolic proteins p40phox, p47phox, and p67phox, and a small GTPase, Rac1 or Rac2 (1). The phagocytic NADPH oxidase complex transfers electrons from NADPH to oxygen to produce reactive oxygen species (ROS), which are released by neutrophils and macrophages to combat pathogens (2,3). A phosphorylation cascade drives NADPH oxidase complex activation, and p67phox contributes to this cascade by regulating phosphorylation of several sites on p47phox (4). In chronic granulomatous disease (CGD), in which patients have mutations in phagocytic NADPH oxidase complex genes, this cascade is disrupted, and the capacity of macrophages and neutrophils to produce ROS is severely impaired (5,6).
