    Background

    Polycomb group RING finger homologs (PCGF1-6) are essential proteins for the assembly of the polycomb repressive complex 1 (PRC1) group of complexes. RING1A/B and a unique PCGF protein make up the catalytic core of PRC1 to ubiquitinate histone H2A. Differences in binding specificities give PRC1 complexes unique functions, depending on which PCGF is included (1-3). Polycomb group RING finger protein 2 (PCGF2), also known as Mel-18, is widely expressed in human tissues and primarily functions as a transcriptional repressor. This protein is broadly expressed in human tissues and has been implicated in numerous regulatory pathways that include cell proliferation and senescence, X chromosome inactivation, embryogenesis, hematopoiesis, and tumor suppression (4-10).

    Furthermore, this protein is known to harbor posttranslational modifications, including phosphorylation and ubiquitination, and is also reported to dimerize (11,12).
