    PPT1 (F7C5U) Rabbit Monoclonal Antibody recognizes endogenous levels of total PPT1 protein. Mouse reactivity is determined by IHC. Nonspecific diffuse cytoplasmic staining was observed in normal human esophageal epithelium and cytoplasmic/nuclear staining was observed in mouse and human small intestine goblet cells.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly250 of mouse PPT1 protein.

    Background

    Palmitoyl-protein thioesterase 1 (PPT1) is a depalmitoylation enzyme that removes palmitate groups from S-acylated proteins (1). PPT1 localization has been demonstrated in multiple compartments, including membrane rafts, the nucleus, lysosomes, the Golgi apparatus, synaptic vesicles, axons, and the endoplasmic reticulum (ER) (1,2). Mutations of the PPT1 protein can cause neuronal ceroid lipofuscinosis (NCL) type 1 (CLN1), a lysosomal storage disorder characterized by brain atrophy, mental and motor retardation, seizures, and retinal degeneration, ultimately resulting in blindness (2,3). In healthy, aged brains, PPT1 and other CLN proteins have been identified as lipofuscin constituents (4).
