    Background

    RBM14, also known as coactivator activator (CoAA), is an RNA-binding protein (RBP) first characterized as a thyroid hormone receptor-binding protein (TRBP) coactivator, with two conserved RNA recognition motifs and a TRBP-interacting domain (1). RBM14 is a capable coactivator that can promote transcription at hormone-response elements and influence alternative splicing (1-3). The RBM14 transcript itself is alternatively spliced, including a shorter, dominant negative form lacking the TRBP-interacting domain, and is commonly referred to as CoAM. This shorter splice variant is upregulated during stem cell differentiation to help control splicing (4,5). The RBM14 protein plays major roles in regulating genomic integrity by influencing many processes, including differentiation, transcription, DNA damage, cell cycle, and condensate formation. RBM14 can function as a repressor by inhibiting the interaction of transcription factors with DNA (6). During non-homologous end joining, RBM14 can recruit XRCC4 and XLF to chromatin and release KU proteins, facilitating DNA repair (7,8). The role of RBM14 in DNA repair has been linked to radiation resistance in glioblastoma (9). In prostate cancer, RBM14 can drive prostate-specific antigen expression even under androgen deprivation (10).
