    RNASEH2B (F5W8X) Rabbit Monoclonal Antibody recognizes endogenous levels of total RNASEH2B protein. This antibody does not cross-react with RNASEH2A protein. This antibody detects a 30 kDa band of unknown identity in some cell lines.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val303 of human RNASEH2B protein.

    Background

    The RNase H2 complex forms as a heterotrimer composed of RNASEH2A, RNASEH2B, and RNASEH2C. RNASEH2A is the catalytic component and is well conserved across species, while RNASEH2B and RNASEH2C are quite divergent (1). The RNase H2 complex is responsible for resolving DNA/RNA hybrids, known as R-loops, which can occur during DNA replication and repair (1,2). The C-terminus of RNASEH2B is bound to PCNA, which then guides the complex to nuclear replication foci to resolve misincorporated ribonucleotides (2). Replicative stress leads to the increased expression of RNase H2 subunits, thereby reducing replication fork stalling in response to chemotherapeutics (3). Mutations in RNase H2 subunits cause Aicardi-Goutieres syndrome (AGS), a rare form of encephalopathy that mimics viral infection due to increased innate immune response from unresolved R-loops (4-6). RNASEH2B is frequently lost along with RB1 in advanced prostate cancer, which sensitizes cells to PARP and ATR inhibitors (7,8).
