    Background

    Septins are a conserved family of GTP-binding proteins. Septin-6 (SEPT6) is a cytoskeletal protein that is expressed in all tissue types, but shows high expression in lymphoid tissues (1). In the brain, it plays a crucial role in regulating neuronal shape, structure, and development, particularly by organizing the dendritic cytoarchitecture. It is highly expressed in neurons, forming complex, ring-like structures at the base of dendritic spines and along axonal branch points, which act as diffusion barriers for membrane proteins (2,3). Septin-6 has become a target in neurodegenerative diseases like Alzheimer's disease (AD). It helps stabilize septin filaments at the cell cortex; when disrupted by pathological tau proteins, it leads to increased, abnormal calcium influx (4).
