    Background

    Lysine methylation is a dynamic posttranslational modification that regulates protein function and various biological processes. SET domain-containing protein 6 (SETD6) is a mono-methyltransferase that promotes the methylation of multiple proteins involved in the regulation of gene expression programs (1). Unlike other lysine methyltransferases (KMTs), SETD6 was first discovered as a non-histone KMT and has been shown to methylate key regulatory factors such as RELA, TWIST1, and BRD4 (2-4). SETD6 can exist in monomer, oligomer, and higher-molecular-weight aggregate forms and its auto-methylation activity is key in stabilizing oligomerization and enzymatic activity (5). Moreover, SETD6 expression is misregulated in a number of cancers. Namely, SETD6 protein expression is upregulated in lung adenocarcinoma tumors, several bladder cancer cell lines, oral squamous cell carcinoma, and in samples from glioma patients (3,6-8).
