    Background

    SIGIRR (Single Ig IL-1-related receptor), also known as IL-1R8, belongs to the IL-1 receptor family characterized by the presence of a common intracellular Toll-IL-1 resistance (TIR) domain and extracellular Ig-like domains (1-3). SIGIRR is widely expressed in several epithelial tissues such as liver, lung, and kidney, and in leukocytes including monocytes, T and B lymphocytes, dendritic cells, and natural killer (NK) cells. It negatively modulates the activation upon ligand binding of certain TIR-containing receptors including members of IL-1 receptor family and TLR family (4-6). Recently SIGIRR was identified as a crucial checkpoint receptor in NK cells and a potential therapeutic target in several pathological conditions (7,8).
