    Background

    Msx2-interacting protein (SPEN or split ends), also known as silencing mediator for retinoid or thyroid hormone receptors (SMRT)/histone deacetylases (HDACs)-associated repressor protein (SHARP), is a potent transcriptional repressor that contains four highly conserved RNA recognition motifs (RRMs) at its N-terminus and an SPOC domain within its C-terminus that is responsible for protein-protein associations with repressive complexes (1,2). SPEN/SHARP has been studied in the context of various cell signaling events, such as the NOTCH or nuclear hormone receptor signaling pathways, and is also critical in the X chromosome inactivation (XCI) process (3-6). More specifically, this protein functions through interacting with X inactive specific transcript (XIST), a long noncoding RNA that primarily functions to inactivate one X chromosome in female cells to achieve dosage compensation. This is achieved by recruiting various repressor complex members, such as SMRT, nuclear receptor corepressor (NCoR), and nucleosome remodeling deacetylase (NuRD) complexes (4-9). SPEN/SHARP has been reported as a novel tumor suppressor that may be clinically utilized as a biomarker for tamoxifen response in ERα-positive breast cancers (10) and has also been shown to be required for various developmental processes (11).
