    Protocol

    Specificity / Sensitivity

TIFA Antibody recognizes endogenous levels of total TIFA protein.

    Species Reactivity:

    Human

    Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro159 of human TIFA protein. Antibodies are purified by peptide affinity chromatography.

    Background

TRAF-interacting protein with FHA domain-containing protein A (TIFA) is a cytosolic adaptor protein that is a critical node in an innate immune signaling axis that is activated upon detection of proinflammatory stimuli derived from a bacterial pathogen-associated molecular pattern metabolite, ADP-heptose (1-4). ADP-heptose binds and activates Alpha-protein kinase 1, which drives the phosphorylation-dependent oligomerization and activation of TIFA (5,6). Activated TIFA is then poised to shape the innate immune response by triggering TRAF6 oligomerization, IKK activation, and NF-κB-dependent proinflammatory gene transcription (1,2,5,7).
